چکیده
It is well known that activin-treated presumptive ectoderm, which is generally called as animal cap, in amphibian differentiates into various types of tissues and organs such as notochord, muscle, heart, and kidney. However, there has been no report that primordial germ cells are induced by activin treatment for animal caps. Anterior or posterior axial mesoderms can be, respectively, induced by middle-low or -high concentrations of activin treatments for dissociated animal cap cells. It is reported that sandwich of two different reaggregates from these dissociated animal cap cells has a potent activity of convergent extension. Using combination of this assay and microarray gene analyses, we tried to isolate new regulators of convergent extension phenomenon. As a result, novel candidate molecule was not isolated from this assay although only a few convergent extension markers were confirmed. Unexpectedly, most genes showing big differences of expression levels in this screening were primordial germ cell markers such as germs, Xpat, and DEADSouth RNA helicase. In addition to them, some novel or functionally unreported molecules were contained in the other high-scored genes of this screening. This is the first report that activin treatment for animal cap cells can induce primordial germ cells, suggesting that the sandwich assay that we used in this study is able to be a good strategy to isolate genes related to primordial germ cell formation.

The role of the desert hedgehog signaling pathway during degeneration and regeneration of peripheral nerves
The desert hedgehog (Dhh) signaling pathway is involved in the development of peripheral nerves (PNs). Dhh-null mice show abnormal neuronal development and perineurial barrier function. As it was previously shown that dhh is mainly expressed in developmental nerves and Sonic hedgehog protein (dhh homologous) has therapeutic effects in neuronal survival, we attempted to investigate the possible ...

متن کاملDynamics of sonic hedgehog signaling in the ventral spinal cord are controlled by intrinsic changes in source cells requiring sulfatase 1.
In the ventral spinal cord, generation of neuronal and glial cell subtypes is controlled by Sonic hedgehog (Shh). This morphogen contributes to cell diversity by regulating spatial and temporal sequences of gene expression during development. Here, we report that establishing Shh source cells is not sufficient to induce the high-threshold response required to specify sequential generation of ve...
متن کاملVentral neural progenitors switch toward an oligodendroglial fate in response to increased Sonic hedgehog (Shh) activity: involvement of Sulfatase 1 in modulating Shh signaling in the ventral spinal cord.
In the embryonic chick ventral spinal cord, the initial emergence of oligodendrocytes is a relatively late event that depends on prolonged Sonic hedgehog (Shh) signaling. In this report, we show that specification of oligodendrocyte precursors (OLPs) from ventral Nkx2.2-expressing neural progenitors occurs precisely when these progenitors stop generating neurons, indicating that the mechanism o...

Gastric cancer (GC) is considered as one of the most serious cancers, and in Iran, due to environmental factors in different regions, it has a high frequency. Apart from environmental factors, genetic and epigenetic ones also play a key role in the development of carcinogenesis of GC. In this regard, the study of functioning of the molecular mechanisms involved in the carcinogenesis and also tu...
